Abstract:OBJECTIVE To prepare phloretin ethosomes and examine its feasibility as a transdermal delivery carrier for phloretin in vitro. METHODS Phloretin ethosomes were prepared by ethanol injection.Encapsulation efficiency and particle size were employed as parameters.Orthogonal test was utilized for optimizing the dosage of phospholipids,cholesterol,ethanol as well as temperature of water bath.Particle size distribution,polydispersity index(PDI)and Zeta potential of ethosomes were determined.Franz diffusion cells were utilized for comparing skin penetration properties of phloretin and its ethosomes. RESULTS The optimal formulations were phospholipids 220 mg,cholesterol 14 mg and ethanol 5 mL with a temperature of water bath 40℃.Encapsulation efficiency,drug loading,average particle size,PDI and zeta potential of phloretin ethosomes were(83.09±1.24)%,(4.86±0.89)%,(162.19±5.88)nm,(0.067±0.011)and(-15.09±2.16)mV respectively.The cumulative permeated drug quantity per unit area of phloretin ethosomes was boosted 3.43 times and permeation process was closer to zero-order kinetics.CONCLUSION The prepared ethosomes have the advantages of high encapsulation efficiency and uniform particle size,and could enhance permeation rate and accumulative penetration of phloretin.Further researches are required.
