Abstract:OBJECTIVE To explore the effect of total glucosides of paeony (TGP) plus glycyrrhizic acid on the expression of inflammatory factors in brain-gut axis of rats with diarrheal irritable bowel syndrome (IBS).METHODS A rat model of diarrhea-type IBS was established by a high-fat diet and ice-water bath stimulation. TGP and glycyrrhizic acid and drug combination groups were assigned. The mechanical pain threshold and intestinal propulsion changes of each group were examined. Then inflammatory pathological changes of prefrontal cortex and jejunum tissue of each group were observed and the effects of interleukin-6 (IL-6), tumor necrosis factor alpha (TNF-α), substance P (SP) and 5-hydroxytryptamine (5-HT) detected.RESULTS Glycyrrhizic acid could significantly boost the mechanical pain threshold of model rats and TGP slowed down the intestinal advancement rate at the same time. After combined use, it markedly blunted the IL-6/TNF-α response of cortex and small intestine tissue and lessoned the brain and intestine tissue Inflammatory response.CONCLUSION TGP plus glycyrrhizic acid have varying degrees of intervention on inflammatory factors in diarrhea-type IBS rats and brain and bowel tissues and combined effect is greatly enhanced. The mechanism of action may be correlated with the releases of internal and intestinal inflammatory factors IL-6 and TNF-α.
