颐脑解郁方对脑出血后抑郁大鼠脑内PI-3K、Akt信号转导通路的影响

摘    要:目的:观察脑出血后抑郁大鼠脑内PI-3K、Akt的表达及中药颐脑解郁方的干预作用。方法:建立脑出血后抑郁模型,采用蛋白印迹(Western-blot)实验方法,研究PI-3K、Akt在脑出血后抑郁模型大鼠脑内左前额叶皮质、海马的表达及中药颐脑解郁方的干预作用。结果:模型组大鼠海马、前额皮质的PI-3K、Akt表达明显高于正常组(P<0·01),中药颐脑解郁方干预后可明显下调PI-3K、Akt在脑内的表达(P<0·01)。结论:卒中后抑郁模型大鼠脑内PI-3K、Akt表达升高,中药颐脑解郁方可使之明显下调,中药颐脑解郁方的抗抑郁作用可能与下调PI-3K/Akt信号转导通路有关。

Abstract:Objective:To observe the expression of PI-3K,Akt and interference effect of yinaojieyu Recipe on the rat model of post-cerebral hemorrhage depression(post-stroke depression,PSD).Methods:Western-blot was used in the rat model of post-stroke depression.The expression of PI-3K,Akt in prefrontal cortex and hippocampus,and the interference of Yinaojieyu Recipe were observed.Results:The expression of PI-3K and Akt in hippocampus and prefrontal cortex in groups of the model of PSD and the false surgery PSD increased obviously compared to that of the normal group.Yinaojieyu Recipe reduced obviously the expression of PI-3K,Akt.Conclusion:The expression of PI-3K and Akt in the brain of PSD rat model increased,while Yinaojieyu Recipe can reduce this expression.The result showed that the anti-depression effect of Yinaojieyu Recipe may be related to its function of reducing expression of PI-3K/Akt signal transduction.
