强痛定联合对乙酰氨基酚对神经病理性疼痛大鼠模型促炎因子的影响

摘    要:目的观察强痛定联合对乙酰氨基酚治疗神经病理性疼痛大鼠促炎症细胞因子肿瘤坏死因子-α(TNF-α)、白介素-1(IL-1)和前列腺素E2(PGE2)的影响。方法 40只SD大鼠随机分为5组:假手术组、坐骨神经慢性压迫模型(CCI)组、对乙酰氨基酚组、强痛定组和联合组(n=8)。手术前1天和手术后第1,3,5,7天测定各组动物的机械性痛阈和热痛阈。采用酶联免疫吸附实验方法检测脊髓TNF-α、IL-1和PGE2浓度变化。采用t检验和方差分析法进行结果分析。结果 CCI可导致大鼠机械性痛阈和热痛阈明显降低,CCI组脊髓TNF-α(3.25±0.34)pg/mg]、IL-1(15.36±1.49)pg/mg]和PGE2(162.57±13.63)pg/mg]升高。与CCI组比较,两种药物联合治疗后能明显改善CCI大鼠痛敏状态,差异均有统计学意义(均P〈0.05),并显著降低TNF-α(1.58±0.27)pg/mg]、IL-1(7.61±0.85)pg/mg]和PGE2(95.16±8.74)pg/mg]的表达,差异均有统计学意义(均P〈0.05)。结论两种药物联合抑制炎症细胞因子释放可减轻CCI大鼠的疼痛反应。

Effects of Bucinperazine combined with Acetaminophen pro-inflammatory cytokines in chronic neuropathic pain rats

Abstract:Objective To observe the effect of Bucinperazine combined with acetaminophen on the expression of Proinflammatory cell tumor necrosis factor-α(TNF-α, interleukin-1 (IL-1) and prostaglandin E2 (PGE2) in chronic con striction injury (CCI) rats. Methods Forty SD rats were randomized into sham operation (sham), CCI (model), Bucin- perazine, Acetaminophen and combined groups (n = 8). PWTL and PWMT were measured before operation and on the 1st, 3th, 5th, 7th postoperative day. The expression of spinal TNF-α, IL-1 and PGE2 were detected by ELISA. t-test and variance analysis methods were used. Results Both mechanical and thermal pain thresholds decreased considerably in CCI group rats. The expression of spinal TNF-α(3.25 ±0.34)pg/mg], IL-I (15.36±1.49)pg/mg], PGE2 (162.57±13.63)pg/mg] increased in CCI group rats. Comparison with CCI group, Bucinperazine combined with acetaminophen group increased markedly (P 〈 0.05). The expresed of spinal TNF-α (1.58±0.27)pg/mg], IL-1 (7.61 ± 0.85)pg/mg], PGE2 (95,16±8.74)pg/mg] decreased significantly (P 〈 0.05). Conclusion The combination of the two drugs can inhibit release of inflammatory mediators and can reduce pain response in CCI rats.
