人T细胞亚群的微量全血染色法

摘    要:本研究建立了人T细胞亚群的微量全血染色法,观察了红细胞溶解液、血细胞与OKT McAb共育时间、洗液及OKT McAb用量等因素对染色率的影响。本法只需0.4ml全血即可得到CD_3、CD_4、CD_8亚群的百分率,同时避免了传统的Ficoll分离法用血量多、淋巴细胞部分选择性丢失等缺点。

Abstract:A method of analysing human T lymphocyte subpopulations by using a micro-quantity of the whole blood was established uponflow cytometer. The experimental conditions, suchas the effects of erythrocyte-lysing solutions. the time of incubation of blood cell with OKT McAb,the quantity of OKT McAb used and etc were observed.It was found that the level of OKT 3, OKT 4 and OKT 8 subpopulations in hamanperipheral blood lymphocytes could be obtained by using only 0.4ml of the whole blood and the staining rate of lymphocytes was found to be increased also. Shortcomings for conventional analysing methods, such as more blood using, partial1y losing of lymphocytes, and so on, could be avoided by using this method.
