TUNEL法检测ER(-)乳癌细胞凋亡和Ki-67表达的研究

目的：探讨雌激素受体阴性乳癌细胞中Ki-67与细胞凋亡之间的关系。方法：通过1．25（OH）2D3诱发雌激素受体阴性乳癌细胞株MDA－MB－435s凋亡，采用MTT法检测细胞增殖情况，TUNEL法检测细胞凋亡指数，免疫组化法检测Ki-67阳性表达指数，进行相关性分析。结果：在10^-8mmol/L 1.25(OH)2D3诱发MDA－MB－435s凋亡过程中，肿瘤细胞 凋亡指数与Ki-67阳性表达指数呈负相关。结论：对凋亡指数及Ki-67阳性表达指数的综合分析，可全面反映细胞凋亡和增殖的平衡状态，对于判断肿瘤细胞的增生具有参考价值。

Study of the expression of Ki-67 and apoptosis tested by TUNEL in the ER(-) breast cancer cell line

Objective: To test the relationship between Ki-67 index and apoptosis in the ER(-) breast cancer cell line. Methods: Apoptosis was induced by 1,25(OH)2D3 in the ER(-) breast cancer cell line. We compared the cell number by MTT, determined quantitatively apoptosis cells by TUNEL, and detected the expression of Ki-67 index immunohistochemically. Results: The expression of the Ki-67 index was decreased during apoptosis that induced by 10-8mmol/L 1,25(OH)2D3 in MDA-MB-435s cell line. there was negative correlation between the apoptosis and the expression of Ki-67 index. Conclusion: A comprehensive analysis of the apoptosis and Ki-67 index reflected the balance of proliferation and apoptosis.
