慢性乙型肝炎患者外周血Th17及相关细胞因子表达研究

目的探讨Th17细胞及相关细胞因子与慢性乙型肝炎(CHB)病情的关系。方法采用RT-PCR检测慢性乙型肝炎轻度、中度、重度患者及健康对照外周血单个核细胞(PBMCs)中Th17细胞的特异性转录因子RORC mRNA及相关细胞因子TGF-β、IL-23、IL-6、IL-22、IL-1βmRNA表达水平,采用ELISA法检测CHB患者及健康对照PBMCs培养上清液中IL-17、IL-22的水平,比较不同组各生物指标分布的差异。结果与健康对照组相比,CHB患者PBMCs中RORC、TGF-β、IL-23、IL-17、IL-6、IL-1βmRNA含量随病情加重而升高,IL-22mRNA随病情加重而降低。CHB患者PBMCs培养上清液中IL-17、IL-22水平,与健康对照组比较,差异均有统计学意义(P均<0.01)。结论 Th17细胞以及诱导Th17分化相关的细胞因子参与了CHB病情发展。

The expression of Thl7 Cells and Thl7 - related factors in peripheral blood of patients with chronic hepatitis B

Abstract ：Objective To study the relationship between the expression of Th-17 cells , ThlT related factors and the activity of the disease in patients with Chronic Hepatitis B（CHB）. Methods Concentration of IL-17, IL-22 in the PBMCs culture supernatant and the expression of RORCmRNA, multiple Thl7 related factors such as IL- 22, IL- 23, IL- 6, IL- 1β, TGF-β were determined with ELISA and RTPCR respectively in 72 patients with CHB （21 patients with mild symptoms,27 pa tients with moderate symptoms,24 patients with severe symptoms and 23 healthy controls（HC））. Results The RORC, IL -23, IL-6, IL-1β, TGFβmRNA expression and IL-17 concentration in the stimulated PBMCs culture supernatant were significantly higher in the patients with CHB than those in controls （ P〈0.01 ） ,while the IL-22mRNA expression and IL-22 concentration were significantly lower in the patients with CHB than those in controls（P〈 0.01 ）. RORC, IL23, IL-6, IL-β,TGF-β expression increased and IL-22 decreased as the disease improved. Conclusion Th17 cells and Thl7related factors might play important roles in the activation of the disease in patients with CHB.
