人参皂苷Rb_1与Rg_1对肾小管细胞缺氧复氧损伤模型的影响

摘    要:目的:探讨人参皂苷的2种单体成分Rb1、Rg1在肾小管细胞缺氧复氧(HRO)损伤模型中的作用及机制。方法:将大鼠肾小管上皮细胞分为对照组、HRO组、Rb1+HRO组和Rg1+HRO组。除对照组外,其余各组给药同时缺氧复氧培养36h后分别检测肾小管细胞增殖活性、增殖细胞核抗原(PCNA)阳性表达情况、丙二醛(MDA)含量、乳酸脱氢酶(LDH)漏出量和超氧化物歧化酶(SOD)活性。结果:人参皂苷Rb1、Rg1可促进HRO性肾小管细胞增殖(P<0.01),增强细胞中PCNA阳性表达(P<0.05),减少肾小管细胞MDA生成及LDH释放,增强SOD活性。结论:人参皂苷Rb1和Rg1对HRO性肾小管细胞损伤模型具有保护作用。

Abstract:OBJECTIVE:To explore the effects and the mechanisms of the two monomers of Ginsenosides(Rb1,Rg1)on cultured rat proximal tubular epithelial(NRK52E)cells with hypoxia-reoxygenation(HRO).METHODS:NRK52E cells were randomly divided into control group,HRO group,(Rb1+HRO)group and(Rg1+HRO)group.After cells were cultured in Rb1 or Rg1 with HRO for 36 hours,the proliferative activity of cells,the content of malondialdehyde(MDA)and the leakage of lactate dehydrogenase(LDH)in culture medium,the activity of superoxide dismutase(SOD)and the expression of proliferating cell nuclear antigen(PCNA)in the cells were respectively detected.The morphology of cultured cells was also observed under phase contrast microscope.RESULTS:Rb1 and Rg1(5mg·L-1)both resulted in decreased content of MDA and leakage of LDH(P<0.01),increased activity of SOD and proliferative activity of cells(P<0.01)as well as increased expression rate of PCNA(P<0.05).CONCLUSION:Rb1 and Rg1 have protective effects on renal tubular cells injured by hypoxia-reoxygenation.
