基于Excel软件的一室模型静脉推注给药方案设计

摘    要:目的:建立一种简便的一室模型静脉注射给药方案设计方法。方法:采用Excel软件编写与给药方案相关的各种数值的计算程序。结果:输入基本参数ke,Vd,以及给药间隔tB、最低有效浓度(MEC)或最小中毒浓度(MTC)后,单变量求解获得最大给药剂量dmax(或最低有效剂量dmax);输入选定的维持量、给药周期n后,电子表格显示第n周期(或稳态)第s次给药后的瞬时血药浓度、达坪分数和负荷剂量,并采用单变量求解获得给药间隔内的有效血药浓度累积时间(T∞)。结论:设计方法简单,使用方便,既能为临床用药提供安全有效的剂量,又能对某一用药方案作出评价。

Dosage Regimen Design of Multiple Dosing Intravenous Administration of One-compartment Model Based on Microsoft Excel
Abstract:Objective: To establish a convenient method of dosage regimen design of multiple dosing intravenous administration of one-compartment model. Methods: Microsoft Excel was used to edit a program. Results: The maximum dose and minimum dose of the administration were obtained after inputting parameters ( k e and V d, ect.), dosing interval ( t s), MEC and MTC by single variable solution in Microsoft Excel tool. After inputting maintenance dose and cycle number (n), the electronic table displayed plasma concentration at any time after the administration, fractional number of reaching plateau and loading dose. The time of the effective plasma concentration in t s or a cycle interval was obtained by single variable solution. Conclusion: This design method can provide a safe and effective dosage for clinical administration and can evaluate a dosage regimen.
