氯沙坦对糖尿病大鼠肾小管间质p38MAPK表达的影响

目的 研究血管紧张素Ⅱ-Ⅰ型受体拮抗剂氯沙坦对糖尿病大鼠肾小管间质p38丝裂原活化蛋白激酶(p38mitogen-actvated protein kinase,p38MAPK)表达的影响.方法 建立链脲佐菌素诱导的糖尿病大鼠模型.设正常对照组、糖尿病组和氯沙坦治疗组,用免疫组织化学和RT-PCR方法观察肾小管间质p38MAPK蛋白及其mRNA表达.结果 p38MAPK在糖尿病大鼠肾小管间质的表达较正常对照组显著升高(P＜0.05),氯沙坦治疗组与同期糖尿病组相比明显降低(P＜0.05).结论 氯沙坦可通过降低糖尿病大鼠肾小管间质p38MAPK表达来缓解肾小管间质病理改变,发挥肾脏保护作用.

EFFECT OF LOSARTAN ON THE EXPRESSION OF p38MAPK IN THE TUBULOINTERSTITIUM OF DIABETIC RATS

Objective To evaluate the effects of Losartan on the expression of p38 mitogen-actvated protein kinase(p38 MAPK) in the tubulointerstitium of diabetic rats.Methods Diabetic rats were induced by Streptozotocin.Wistar rats were divided into the control group(NC),the diabetic group(DM) and the diabetic group treated with losartan(DT).The change in the expression of p38MAPK proteins was analyzed with the routine immunohistochemical method.At the same time,expression of p38 MAPK mRNA was detected by RT-PCR.Results Immunohistochemical staining and RT-PCR showed that the level of p38 MAPK of group DM was significantly higher than that of group NC(P<0.05).The level of p38 MAPK of group DT was lower than that of group DM(P<0.05).Conclusion Losartan could inhibit the over-expression of p38 MAPK mRNA and protein in the tubulointerstitium and prevent the progress of the tubulointerstitial injure in STZ induced diabetic rats.
