单克隆抗体—重氮胶乳作心钠素大鼠组织定位

摘    要:作者用重氮胶乳为载体,与心钠素单克隆抗体(McAb)形成McAb-重氮胶乳,用免疫重氮胶乳凝集实验,对10只Lou/c大鼠不同组织和器官的心钠素浓度进行了定性分析,证实心房提取液凝集反应最强(?),其次为肾脏(?)。同时用心钠素Ⅲ-McAb的放射免疫分析法(RIA),进行定量分析。心房为1919.0ng/g组织湿重,肾脏为22.0ng/g组织湿重,心肌为20.0ng/g组织湿重,脑为76.1ng/g组织湿重,与定性分析结果基本相似。证实这种免疫重氮胶乳可以对组织和器官中的心钠素进行初步定位研究,也可用于McAb细胞株的筛选和鉴定,实验结果快速、可靠。

Abstract:Diazo-latex as a carrier was coupled with cardionatrin monoclonsl antibody (McAb) and McAb-diazo-latex was produced. Cardionatrin concentration in different tissues and organs of 10 Lou/c rats was qualitatively analysed by immuno-diazo-latex ,agglutination testing. Our results show that the agglutinative reaction of the atrial extracts was the strongest ,the next strongest being one reaction of the kidney extracts . At the same time, all the tissue extracts were quantitatively analysed with RIA of cardionatrin-McAb. The atrium was 1919 ng/g tissue wet. The kidney, myocardium and brain were 22 ng/g tissue wet,20 ng/g tissue wet,76 ng/g tissue wet, respectively. The cardionatrin concentration of the lung, spleen and muscle was the lowest. These results were similar in many ways to those produced by qualitative analysis and suggest that immuno-diazo-lalex is useful for the study of cardionatrin localization in different tissues and organs and for the screening and identification of McAb hybridoma This is a rapid and rcliablc method.
