浅低温联合硫酸镁治疗对全脑缺血大鼠的脑保护作用研究

摘    要:目的:探讨浅低温联合硫酸镁治疗对大鼠全脑缺血再灌注后的神经保护作用及其与钙结合蛋白表达的关 系。方法:30只SD大鼠随机分为四组:浅低温组(8只),硫酸镁组(8只),浅低温联合硫酸镁组(8只),对照组(6只)。 采用窒息+艾司洛尔(超短效β受体阻滞剂)的心肺骤停模型使大鼠出现全脑缺血。7d后大鼠处死、取脑、切片,免疫组 化分析钙结合蛋白D28k(CB)的活性表达情况,透射电镜下观察脑组织损伤情况,组间数据比较使用t检验和非参数 Kruskal-Wallis检验。结果:浅低温联合硫酸镁组神经元中钙结合蛋白表达程度明显高于对照组(P<0.01),也好于其 它2个单独治疗组(P<0.05),电镜下观察其海马区神经组织损伤程度也较轻。结论:浅低温联合硫酸镁治疗可以减轻 脑缺血再灌注损伤,并好于浅低温或硫酸镁单独治疗,机制可能与促进钙结合蛋白表达有关。

Abstract:Objective: To investigate the neuroprotective effect of modest hypothermia combined with magnesium sulfate on rat brain after transient global ischemia and the influence on expression of calbindin D28k. Methods: 30 Sprague-Daw-ley rats were randomly divided into four groups: modest hypothermia (8 rats) , magnesium sulfate (8 rats) , modest hypothermia combined with magnesium sulfate (8 rats), control (6 rats). Cardiac arrest (CA) model was founded. We obtained the brain slices after 7 days and calculated calbindin D28k(CB)expression by immunochemistry methods, and observed histological damage under electron microscope. Results: CB expression of modest hypothermia combined with magnesium sulfate groups was more than that of control group (P<0. 01) and the other two groups (P<0. 05), and their cerebral histological damage degree were also lighter than other groups. Conclusion: Modest hypothermia combined with magnesium sulfate treatment can effectively alleviate damage of cerebral ischemia/reperfusion probably by promoting the expression of CB, and it is more effective than either treatment used alone.
