硫酸镁对实验性脑出血大鼠细胞凋亡及Caspase-3表达的影响

摘    要:目的研究硫酸镁对实验性脑出血大鼠模型神经细胞凋亡及Caspase-3表达的影响。方法将108只大鼠随机分为3组:假手术组、模型组和硫酸镁干预组各36只,应用立体定向技术将自体不凝血注入大鼠尾状核区制备脑出血模型,制模成功后用25%硫酸镁400 mg/kg腹腔注射,分别在术后1d、3d、7d、14d、21d、28d不同时间点各取6只大鼠断头取脑,连续切片作TUNEL染色和Caspase-3免疫组化染色。结果与假手术组比较,模型组脑组织中凋亡的神经细胞及Caspase-3表达均增加(P<0.05),在使用25%MgSO_4治疗后干预组神经细胞凋亡明显减少,Caspase-3表达降低,随着用药时间的延长,脑组织中神经细胞凋亡及Caspase-3表达均较模型组有明显降低(P<0.05)。结论硫酸镁可通过抑制脑出血大鼠神经细胞凋亡与Caspase-3活化从而起到显著的脑保护作用,且随应用时间的延长疗效更加显著。

The effect of magnesium sulfate on the expression of Caspase-3 in experimental intracerebral hemorrhage rats

Abstract:Objective To study the effects of magnesium sulfate on apoptosis and the expression of Caspase-3 in intracerebral hemorrhage(ICH)rats.Methods 108 SD rats were randomly divided into control group,hemorrhage group and magnesium group.ICH was induced in rats by stereotactic infusion of in-coagulable blood 50μl into the caudate nucleus,25% magnesium sulfate(400 mg/kg)was injected into rats.TUNEL method was used to detect apoptosis,and immunohistochemical method was used to detect the expression of Caspase-3 in cerebral tissues at different time point of 1st day,3rd day,7th day,14th day,21st day and 28th day.Results Compared with sham operation group,the apoptosis and expression of Caspase-3 in ICH rats increased significantly(P<0.05).Caspase-3 expression preceded apoptosis both at the beginning and peaking time.The apoptosis and the expression of Caspase-3 decreased remarkably after treatment with magnesium sulfate. There was no difference between the groups treatment with different dose of magnesium sulfate.Along with time,there were obviously differences of the indexes mentioned above in the other groups compared with the hemorrhage group(P<0.05). Conclusion Magnesium sulfate could significantly restrain the apoptosis and the expression of Caspase-3,which lead to protect the brain tissues.The effect would be more obviously along with the treatment time prolonged.
