胶体金渗滤试验检测乙型肝炎患者血清中IgM抗体的研究

目的建立胶体金免疫渗滤法检测乙型肝炎患者血清中IgM抗体的方法,评价其灵敏度、特异度和重复性。方法以小乙型肝炎表面抗原作为捕获抗体包被硝酸纤维素膜、抗人IgM抗体作为检测抗体标记胶体金,建立金标免疫渗滤法检测乙型肝炎患者血清中IgM抗体。结果 102份乙型肝炎患者血清,抗HBV IgM阳性标本99份,检出率97%。50份正常血清中,有一份检测结果弱阳性,特异度98%。结论胶体金免疫渗滤法检测乙型肝炎IgM抗体敏感性高、特异性强、重复性好,且方便快捷,不需要仪器设备,在乙型肝炎IgM抗体快速检测上有广阔的应用前景。

Detection of Anti-HBsAg IgM in Serum by Immunogold Filtration Assay

Objective To develop immunogold filtration assay(IGFA)for the detection of anti-HBsAg IgM and evaluate its sensitivity,specificity and repeatability.Methods Nitrocellulose membranes were coated with small HBsAg as capture antibody,and anti-human IgM antibody as detection antibody to label immunogold,by which we developed the direct method to detect IgM antibodies in serum of patients with hepatitis B.Results The results showed that 99 out of 102 total cases were positive in serum of patients with hepatitis B.The sensitivity of IGFA was 97%.The specificity was 98% in 50 healthy individuals with one result of weak positive.Conclusion IGFA is featured with high sensitivity,strong specificity and good repeatability and convenience without need for equipment in the detection of HBsAg,where extensive application as a rapid detection tool is expected.
