摘    要:目的 探讨了线粒体CO1基因作为DNA条形码对鼠类进行物种鉴定的可行性.方法 将从中尼边境采集到的42个鼠类标本,提取基因组DNA,用通用引物PCR法扩增线粒体CO1基因,并测序.将测序结果与GenBank中其他鼠类物种的DNA条码进行BLAST比对,并构建分子进化树.结果 本研究中有17个标本能通过PCR扩增出特异性CO1基因条带,17个标本的种内遗传距离平均值为0.035,种间遗传距离平均值为0.47,种间遗传距离是种内遗传距离的10倍以上.根据BLAST比对法,将采集到的17个标本分到不同的鼠种中.结论 CO1基因序列能够对鼠类进行有效的物种鉴定.

Abstract:Objective To discuss the feasibility of using mitochondrial CO1 gene as DNA bar code to identify species of murid. Methods The genome DNA was extracted from 42 murid samples collected in the Sino - Nepali border area. The mitochondrial COl gene was amplified by method of consensus primer PCR and sequenced. BLAST comparison was made between the sequencing results and the DNA bar codes of other murid species in genebank,and molecular evolution tree was built. Results In this research, specific CO1 gene straps in 17 samples were amplified by PCR. The average intraspecies genetic distance of the 17 samples was 0. 035. The average interspecies genetic distance was 0.47. The interspecies genetic distance was almost 10 times more than the intraspecies genetic distance. According to the BLAST comparison,the 17 samples were classified in different murid species. Conclusion The CO1 gene order can be used effectively to identify the murid species.
