摘    要:本文在体外对新生大鼠(0~1d)大脑皮层神经细胞进行原代培养的基础上,建立谷氨酸(Glu)对培养的皮层神经细胞兴奋毒损伤模型。通过检测乳酸脱氢酶释出率和形态学观察,证实了Glu对神经细胞存在兴奋毒作用。通过测定不同条件下Glu兴奋毒对培养的神经细胞造成损伤时细胞膜脂质过氧化物(LPO)浓度,发现膜LPO浓度与Glu剂量及作用时间紧密相关,表明Glu对神经细胞兴奋毒作用有自由基产生并介导了神经细胞的损伤。

Abstract:The model of glutamate(Glu) excitotoxicity in primary cultures of neurons from new-born(o-1d) rat cortex was set up. The excitotoxic damage was assessed by lactate dehydrogenase release test and morphological observation of cultrued neurons. The membrane lipid peroxide(LPO) of damaged neurons in primary cultures was tested under different Glu excitotoxic conditions. It was found that the concentration of membrane LPO was relative to Glu dose or exposure duration. The results suggest that Glu excitotoxicity to neurons proudces free radical and the free radical mediates neuron damage.
