急性脑缺血脑部神经元Fos表达与病理变化的实验研究

摘    要:本研究采用免疫组化染色显示Fos表达来反映急性缺血后大鼠前脑顶叶、海马区和下丘脑神经元的功能活性,采用尼氏染色、透射电镜观察神经元的病理变化。结扎13只大鼠双侧颈总动脉后2h,顶叶皮质Fos免疫阳性神经元呈密集分布,海马呈中等密度分布,下丘脑呈稀疏分布。光镜下观察尼氏染色的神经元,海马损伤者较多(77.48%),顶叶皮质次之(66.18%),下丘脑较少(50.11%)。电镜观察损伤的神经元粗面内质网、线粒体、高尔基复合体肿胀,核糖体聚集,严重者粗面内质网消失。实验结果提示缺血敏感的神经元Fos表达较强,Fos表达与神经元的缺血损伤明显有关,可能对神经元的缺血损伤起保护作用。

Abstract:By using immunohistochemical straining and transmission electron microscope .neuronal functional activity and potholog-ical changes: were examined in forebrain parietal lobe, hip- pocampal formation and hypothalamus in the rat following a-cute ischemia. Distributions of the Fos - like immunoreactive neurons were dense in the cortex of parietal lobe,middle density in hippocampus,A few scattered in hypothalamus in 2h of ligated common caroited artery in thirteen rats. By light microscope with Nissl straining, neurons of pathological changes were 77. 48% in hippocampus,66. 18% in the cortex of parietal lobe,50. 11% in hypothalamus. By electron microscope injured neurons showed swelling mitochondria, rough endoplas-mic reticulums ribosomes qathering and rough endoplasmic reticulum disappearrance in serious injured neurons. Fos expression is intense in neuron vulnerable to ischemia,correlated with neuronal damage to ischemia.
