ω-芋螺毒素SO3对大鼠慢性神经痛镇痛作用及对血清IL-6浓度影响

摘    要:目的 观察持续鞘内注射新型特异性N型电压依赖性钙通道阻滞剂-ω-芋螺毒素SO3对慢性压迫性损伤所致的神经痛大鼠的长期镇痛效果,及其对神经痛大鼠血清IL-6浓度的影响。方法 建立大鼠CCI模型,比较观察连续鞘内注射ω-芋螺毒素SO3后大鼠热刺激爪退缩阈值和冷水中抬足次数变化以及血清IL-6水平的改变。结果 CCI大鼠术后出现热刺激爪退缩阈值缩短(P〈0.01)、冷水中抬足次数增加(P〈0.01);血清中IL-6水平明显升高(P〈0.01)。持续鞘内注射生理氯化钠溶液对这两种阈值变化以及血清中IL-6水平无明显影响;持续鞘内注射ω-芋螺毒素SO3(60ng/h)能明显提高CCI大鼠热刺激爪退缩阈值(P〈0.01),减少冷水中抬足次数(P〈0.01);并显著降低血清中IL-6水平(P〈0.01)。结论 长期持续鞘内注射ω-芋螺毒素SO3可有效地治疗CCI大鼠的冷、热痛觉过敏,并显著降低CCI大鼠血清IL-6浓度。

Abstract:Aim To evaluate the effect of continuous intrathecal administration of omega-conotoxin SO3 on a model of neuropathic pain and the down-regulation of serum concentration of IL-6 in a rat model of chronic constriction injury.Methods CCI rat models were established.The threshold of thermal and cold allodynia were observed and the serum concentration of IL-6 was measured after continuous i.t.omega-conotoxin SO3.Results In the rats undergoing CCI surgery,withdrawal times to thermal stimulation were significantly exhibited(P<0.01),and with drawal frequencies to cold stimuli were significantly increased(P<0.01).The serum concentration of IL-6 was significantly increased in rats of C group(P<0.01).But there was no difference in cold and thermal stimulation threshold and serum concentration of IL-6 between C group and CN group.Continuous i.t.omega-conotoxin SO3 60ng/h produced a significant increase of withdrawal times to thermal stimulation (P<0.01),and a significant reduction of withdrawal frequencies to cold stimuli (P<0.01).The serum concentration of IL-6 in CS group was significantly decreased after continuous i.t.omega-conotoxin SO3 (P<0.01).Conclusion Continuous i.t.omega-conotoxin SO3 produced significant antinociceptive effects on both cold and thermal hyperalgesia while decreasing the serum concentration of IL-6.
