| 摘 要： | 目的：观察转染PTEN的鳞癌细胞的增殖情况及PTEN在口腔白斑及鳞癌中的表达，探讨PTEN基因在口腔鳞癌发展过程中的作用。方法：采用RT-PCR法从正常组织中扩增PTEN基因，构建真核表达载体pcDNA-PTEN，采用脂质体将pcDNA-PTEN转染人舌鳞癌（Tca8113）细胞系，MTT法观察转染前后细胞的增殖情况，流式细胞术检测PTEN基因转染后细胞各周期细胞所占比例。免疫组织化学技术检测口腔白斑（25例）和口腔鳞癌组织（32例）中PT
EN蛋白表达率。结果：成功构建pcDNA-PTEN载体，并获得阳性转染细胞。与空载体组相比，转染pcDNA-PTEN组的舌癌细胞系Tca8113细胞的生长抑制率具有显著性差异(P<0.01)；与空载体组比较，转染PTEN基因的Tca8113细胞G0-G1期细胞数增多（P<0.01）, S期细胞减少（P<0.01）；PTEN在口腔鳞癌中表达减少或缺失，且与组织分化程度明显相关（P<0.05）。结论：PTEN的表达可抑制舌鳞癌细胞的生长；PETN在舌鳞癌发展过程中发挥抑癌作用。
