摘    要:目的:探讨瘦素(Leptin)对大鼠肝星状细胞(HSC)硬脂酰辅酶A去饱和酶-1(SCD-1)基因表达的影响,为进一步寻找糖尿病人合并脂肪肝的形成机制提供新的思路。方法:将培养好的肝星状细胞系分为两组,分别应用100ng/ml瘦素和DMEM细胞培养液培养,运用RealTimePCR的方法研究瘦素对SCD-1基因的调节作用。结果:应用RealTimePCR技术检测瘦素可以下调SCD-1基因的表达,下调倍数0.315倍。结论:高浓度瘦素可能通过下调SCD-1基因表达参与脂肪肝的形成,可能是导致糖尿病及代谢综合征合并脂肪肝的原因之一。

The effect of leptin on SCD-1 gene expression of the rat hepatic satellite cell

Abstract:Objective:To study the effect of leptin on SCD-1 gene expression of the rat hepatic satellite cell.Method:Two groups(control and experiment group)were culturaled with control solution and leptin respectively.The expression of SCD-1 mRNA in the hepatic satellite cell were detected by RT-PCR.Results:The expression of SCD-1gene were down regulated in the hepatic satellite cell which culturaled with leptin,and its expression was only 0.315 of that in HSCs which culturaled with control solution.Conclusion:SCD-1 gene expression could be down-regulated by leptin,and it suggest that leptin may play an inportant role in the formation of nonalcoholic fatty liver in diabetes or metabolic syndrome by regulating the expression of SCD-1 gene.
