氨胺-T法和Iodogen法碘标记抗体的比较

摘    要:本文比较了氯胺-T法和Iodogen法碘化标记单克隆抗体BAC5在不同反应条件下的标记率,标记反应的时间与抗体免疫活性之间的关系。结果表明:这两种标记方法的最佳pH值为7~8;反应体积在100~200μl范围内均可以得到较好的结果;氯胺-T法反应3min,标记率已接近最大值,而Iodogen法要在10min后才有最大的产额。结果还表明:在本实验条件下,氨胺-T法和Iodogen法的反应时间分别小于3min和5min时,其抗体的免疫活性可保留原来的95%以上。

Abstract:he radioiodination technique has been applied extensively in the isotope labeling of antibodies.This study compared the radiolabeling yields of monoclonal antibody BAC5iodinated by iodongen and chloramine-T methods under different conditions and observed the relationship between the immunoativity and the labeling time.It has beenshown that the maximal yield occurred in the range of pH7-8 both methods,and the optimal reaction time(t)and volume(V)for chloramine-T and Iodogen methods were 3minutes(t), 500ul(V) and 10 minutes(t),between 100 and 300 l(V) respectively.95% of its original immunoactivity has been remained with the labeling time of less than 3 and 5 minutes by chloramine-T and Iodogen methods respectively.
