牛脑提取液对周围神经再生影响的实验研究

摘    要:目的研究牛脑提取液对周围神经再生的影响。方法选用180~200 g SD大鼠48只,切除5 mm长的坐骨神经,两断端用硅胶管桥接形成再生室。局部用药组右侧再生室内注入牛脑提取液(Extract of Calf Brain,ECB),左侧再生室内注入生理盐水(Normal Saline Solu-tion,NS)作对照,观察局部应用EBC对神经再生的影响。全身用药组腹腔内分别注入EBC原液和稀释1倍的EBC及NS作对照,观察全身应用EBC对神经再生的影响。结果①小腿三头肌湿质量:实验组明显高于对照组(P<0.01)。②光镜及电镜观察及轴突计算机图像分析:实验组再生的有髓神经纤维较粗,髓鞘厚,呈板层样排列;轴突计数及直径均优于对照组(P<0.01);有髓神经纤维髓鞘较厚、轴突直径较粗,神经再生活跃。而对照组中有髓神经纤维髓鞘较薄、直径细小。③辣根过氧化物酶标记:实验组脊髓腰段横切片中,可见校多标记的前角运动神经元。而对照组中则未发现。全身用药组与局部用药组结果相似,全身小浓度用药组好于大浓度用药组。结论全身和局部应用ECB对周围神经再生有明显的促进作用。EBC也支持运动神经元存活。选用适宜的ECB浓度效果更好。

Abstract:Objective It is to investigate the effect of extract of calf brain to regeneration of peripheral nerve.Methods 48 SD rats which weight 180g to 200g were used.The sciatic nerve segment length 5 mm was removed,and the regeneration chambers were made by using silicone tube to connect the proximal and distal ends on all rats.Extract of Calf Brain(ECB) was locally or intraperitoneally injected to the rats which were used as experimental group.While the normal saline solution(NS) as control.The influence of ECB on the nerve regeneration was observed.Results ①The gastrocnemiue muscle wet weight were heavier in experimental groups than that in control groups(P<0.01).②The optical and electronic microscopic image analyses showed that there were thicker myelin,thicker axon and more highly vigorous regeneration of nerve fibers in experimental groups than taht in control groups(P<0.01).③The HRP-labelled motor neurons in anterior horn of spinal cord were observed in experimental groups,but not observed in control groups.The results of the intraperitoneally injection groups were similar to that of the local injection group of ECB.The results of the low concentration group were better than that of the high concentration group.Conclusion Both local and intraperitoneally injection of EBC can obviously promote regeneration of injured peripheral nerve.ECB can also support the life of motor neuron.It is much better to use it in proper concentration.
