左归丸、右归丸对老年大鼠海马糖皮质激素受体mRNA表达的影响

摘    要:目的:观察左归丸、右归丸对老年大鼠海马糖皮质激素受体(GR)mRNA表达的影响,探讨海马对下丘脑-垂体-肾上腺(HPA)轴的调节作用,以及左归丸、右归丸延缓老年大鼠神经内分泌调控退的作用机制。方法:以自然衰老大鼠为动物模型,采用原位杂交法,观察药物对大鼠海马各分区(DG、CA1、CA2、CA3)GRmRNA表达变化,结果:老年大鼠海马各分区GRmRNA表达均明显减弱,而左归丸、右归丸能不同程度地提高老年大鼠海马DG、CA1和CA3分区低下的GRmRNA的表达,结论:左归丸、右归丸通过提高GRmRNA表达,进而改善海马对HPA轴的抑制性调控作用,以延缓机体衰老。

Abstract:Object: To study the effect of hippocampus on hypothalamic pituitary adrenal (HPA) axis and the mechanism of how the Chinese traditional medcine Zuogui Wan of yougui Wan to postpone aging. Method: Male SD rats were grouped as follows: (1) young control group; (2) old control group; (3) old group with Zuogui Wan; (4) old group with Yougui Wan. Using in situ hybridization, GRmRNA expression in subfields of the hippocampus were observed. Results: Compred to young control group, the GRmRNA expression of old rats weakened obviously. Meanwhile, the GRmRNA expression of the old group with Zuogui Wan and the old group with Yougui Wan was improved. Conclusion: Aging maybe relate to the imbalance of hippocampus' regulating the HPA axis; Zuogui Wan and Yougui Wan can stave aging through improving the function of hippocampus to the HPA axis.
