滋肾清热活血方治疗大鼠系膜增生性肾炎的实验研究

摘    要:观察滋肾清热活血方对系膜增生性肾炎(MsPGN)的疗效,并探讨其机制。采用SD大鼠系膜增生性肾小球肾炎模型,分正常对照组、模型对照组,治疗组对比研究。观察肾小球系膜区情况及各组大鼠肾小球培养上清中白细胞介素-1(IL-1)的活性等。结果:治疗组与模型对照组比较,肾小球系膜细胞(MC)的增生和系膜基质的积累明显受到抑制,系膜区致密电子物沉积显著减轻;肾小球培养上清中IL-1活性显著降低。提示:滋肾清热活血方能抑制MsPGN系膜区MC的增殖及基质的积累,其作用可能与抑制肾小球分泌的IL-1活性有关。

Abstract:The therapeutic effect of "ZiShenQingReHuoXue" recipe treating mesangial proliferative glomerulonephritis (MsPGN) of rats and its mechanism was explored. A model of rat MsPGN was established. All rats were divided into three groups: normal group, model group and "ZiShenQingReHuoXue" recipe treated group (treatment group). Red blood cells (RBCs) in urine were observed under light microscopy. Serum levels of creatinine and 24 h urine protein were determined, and mesangial cells and matrix in glomeruli of rats were observed under light microscopy and electron microscopy. Mesangium area proportion in glomerular capillaries area was calculated using computed imaging analysis system. Interleukin 1 (IL 1) activity in the supernatant of cultured glomeruli from rats at 8th week was determined by using conventional methods. Results showed that MC proliferation and matrix accumulation in glomeruli were suppressed, electron dense deposits in mesangium were decreased in treatment group as compared with model group. The mesangial area proportion in glomerular capillaries area in model group was significantly higher than normal group and treatment group. IL 1 activity in the supernatant of cultured glomeruli in model group was significantly higher than in normal group and treatment group. It was concluded that "ZiShenQingReHuoXue" recipe could strongly suppress MC proliferation and matrix accumulation in MsPGN of rats, so as to treat MsPGN. Decreased IL 1 activity might contritute to the therapeutic effect of "ZiShenQingReHuoXue" recipe on MsPGN.
