| 摘 要： | 目的探讨体外诱导骨髓间充质干细胞（BMSCs）向膀胱尿路上皮细胞分化的可行性研究,为组织工程膀胱的建立提供一种新的种子细胞来源.方法抽取猪的骨髓液在体外行全血原代培养、扩增及鉴定骨髓间充质干细胞.用机械分离法获取膀胱尿路上皮组织,对其行原代培养、扩增、鉴定并提取尿路上皮细胞培养液.选取第三、四代的BMSCs和尿路上皮细胞分成四组：A组：BMSCs＋尿路上皮细胞＋表皮细胞生长因子;B组：BMSCs＋尿路上皮细胞条件培养液＋表皮细胞生长因子;C组：BMSCs＋表皮细胞生长因子;D组：BMSCs 2 mL.结果用流式细胞仪检测BMSC表面特异阳性标志物CD29、CD44,阴性标志物CD45,结果均为阳性.用角蛋白AE1/AE3单克隆抗体鉴定尿路上皮细胞及实验A、B组结果均为阳性,C、D组均为阴性.结论骨髓间充质干细胞具有多向分化的能力,模拟体内的微环境可使BMSC向尿路上皮细胞分化.
