免疫化疗对口腔鳞癌细胞增殖及凋亡的影响

摘    要:目的 评价白细胞介素—2(IL—2)局部注射联合化疗对口腔鳞癌细胞增殖、凋亡特性的影响。方法 34例T3、T4期口腔鳞癌患者随机分2组接受术前治疗,其中23例行免疫化疗(IL—2局部注射合并PVP化疗),11例行单纯PVP化疗。采用免疫组化和原位DNA末端标记法分别检测治疗前后肿瘤增殖细胞核抗原和细胞凋亡的表达情况,计算出增殖指数和凋亡指数。结果 免疫化疗组治疗后肿瘤细胞增殖指数明显低于单纯化疗组(P<0.05),细胞凋亡指数明显高于单纯化疗组(P<0.05)。经过免疫化疗对抑制口腔鳞癌细胞增殖,诱导细胞凋亡具有重要意义。

Abstract:Objective The effect of locoregional immunotherapy of interleukin 2 (IL-2) in combination with chemotherapy upon cell proliferation and apoptosis was evaluated in oral squamous cell carcinomas.Methods Thirty-four patients with stage T3 or T4 oral squamous cell carcinoma were randomly divided into two groups, and treated with two therapies, 23 cases of whom received immunochemotherapy, and 11 cases, PVP chemotherapy. PCNA and Apoptosis expressions at tumor site before and after therapy were detected with immunohistochemistry and terminal deoxynucleotidyl transferase mediated dUTP-biotin nick end labeling(TUNEL) methods. PCNA index (PI) and Apoptosis index (AI) were the percentage of PCNA-positive cells and TUNEL-positive cells in the tumor cells, respectively. Results There was a significant difference in PI or AI between immunochemotherapy and chemotherapy groups. PI decreased significantly, however,AI increased greatly, after immunochemotherapy.Conclusion Immmunochemotherapy for oral squamous cell carcinomas may play an important role in inhibition of cell proliferation and induction of apoptosis.
