加减葳蕤汤对青霉素致小鼠上呼吸道菌群失调的调节作用

摘    要:目的研究加减葳蕤汤对青霉素致小鼠上呼吸道菌群失调的调节作用.方法通过腹腔注射青霉素建立小鼠菌群失调模型,观察加减葳蕤汤对小鼠口咽部优势菌甲型链球菌的构成比和菌群总密集度的调节作用.实验分4组:(1)阴性对照组;(2)青霉素模型组;(3)阳性药物对照组;(4)不同剂量(高中低剂量)加减蒇蕤汤组.结果(1)小鼠口咽部甲型链球菌构成比和菌群总密集度在腹腔注射青霉素后显著降低,青霉素致小鼠上呼吸道菌群失调模型复制成功.(2)不同剂量的加减葳蕤汤对菌群失调小鼠上呼吸道菌群有不同程度的调节作用.高剂量的加减葳蕤汤能显著升高甲型链球菌的构成比;中剂量的加减葳蕤汤能够显著增进菌群总密集度;低剂量的加减蒇蕤汤对甲型链球菌的构成比及菌群总密集度都有一定的促进作用.结论青霉素致小鼠上呼吸道菌群失调模型可用于中医药调节小鼠上呼吸道菌群失调的研究;加减葳蕤汤对青霉素致小鼠上呼吸道菌群失调具有调节作用,其作用机制可能与促进有益菌甲型链球菌的生长,升高菌群总密集度、促进菌群多样性有关.

Abstract:Objective To explore the regulating effects of Jia Jian Wei Rui Decoction on dysbacteriosis in upper respiratory tract of mice induced by Penicillin. Methods We established dysbaeteriosis model in mice by intraperitoneal injection of Penicillin, and then we observed the regulating effect of Jia Jian Wei Rui Decoction on the ratio of α -hemolytic streptococcus and the total dopulation density of bacteria in the oral pharynx of mice. We devided the mice into four groups: ① negative control group; ② Penicillin model group; ③ positive drug control group; ④ Jia Jian Wei Rui Decoction groups with different dose. Results (1) The ratio of a -hemolytic streptococcus and the total dopulation density of bacteria in the oral pharynx of mice decreased after intraperitoneal injection of Penicillin, Jian Wei Rui Decoction the dysbacteriosis model in mice was established successfully. (2) Different dose of Jia had different regulating effects on micro-ecology in upper respiratory tract of mice with dysbacteriosis. High dose of Jia Jian Wei Rui Decoction increased significantly the ratio of α-hemolytic streptococcus, middle dose of Jia Jian Wei Rui Decoction improved the total dopulation density of bacteria, and low dose of Jia Jian Wei Rui Decoction increased both the ratio of α-hemolytic streptococcus and the total dopulation density of bacteria. Conclusions Penicillin induced dysbacteriosis model in mice can be used in studying the regulating effects of traditional Chinese medicines on dysbacteriosis in upper respiratory tract of mice. Jia Jian Wei Rui Decoction has the regulating effect on dysbacteriosis in upper respiratory tract of mice induced by Penicillin, and the mechanisms may be related with promoting the growth of beneficial α-hemolytic streptococcus, improving the total dopulation density of bacteria, and promoting the diversity of bacteria.
