消瘤方对人子宫内膜干细胞凋亡的影响

目的探讨消瘤方对人子宫内膜干细胞凋亡的影响。方法分离子宫内膜异位症患者在位、异位内膜干细胞,检测干细胞相关标记物CD34、CD117抗原及Oct-4因子、ABCG2蛋白的表达;将分离出的子宫内膜干细胞分为对照组(空白血清+生理盐水)、西药组(空白血清+抑那通)、消瘤方组(消瘤方含药血清+生理盐水)、综合治疗组(消瘤方含药血清+抑那通),予体外药物干预后检测干细胞凋亡率、凋亡相关蛋白Bcl-2、Bax的表达情况。结果 CD34、CD117抗原及Oct-4因子、ABCG2蛋白在子宫内膜干细胞中呈阳性表达;药物干预后,综合治疗组诱导干细胞凋亡优于其他组,各组在位(异位)内膜干细胞的凋亡率分别为对照组0.60%(0.63%)、抑那通组35.20%(36.41%)、消瘤方组38.72%(39.80%)、综合治疗组78.53%(79.60%);分离的Bcl-2、Bax蛋白表达有意义。结论子宫内膜异位症的发生与在位、异位内膜干细胞凋亡能力的下降有关,中药消瘤方联合抑那通具有一定的诱导子宫内膜干细胞凋亡的作用。

Effect of “Xiaoliu Decoction” on apoptosis of endometrial stem cells

Objective To separate and culture the endometrial stem cells and explore the effects of＂Xiaoliu Decoction＂on apoptosis of endometrial stem cells.Methods The endometrial stem cells of the ectopic and vitro endometrium in patients were separated. We detect the expression of stem cells related markers such as CD34,CD117 antigen and Oct-4 factor,ABCG2 protein; the endometrial stem cells were divided into control group（treated with serum and saline）,western medicine group（treated with serum and Enantone）,＂Xiaoliu Decoction＂group（ treated with serum containing ＂Xiaoliu Decoction＂and saline）,combined therapy group（treated with serum containing＂Xiaoliu Decoction＂and Enantone）. The expression of apoptosis,Bcl-2 /Bax protein of endometrial stem cells were detected in vitro. Results CD34,CD117 antigen and Oct-4 factor,ABCG2 protein in endometrial cells took positive expression; after medicine intervention,the apoptosis of endometrial stem cells in the drug intervened group and combined therapy group were better than that in the other groups,The apoptosis rates of ectopic（vitro） endometrium of the control group,western medicine group,＂Xiaoliu Decoction＂group and combined therapy group were 0.60%（0.63%）,35.20%（36.40%）,38.72%（39.80%） and 78.53%（79.60%） respectively; the express of Bcl-2/Bax protein was meaningful. Conclusion The occurrence of endometriosis has relationship with the apoptosis decline of stem cells in vitro or ectopia,Combined＂Xiaoliu Decoction＂and Enantone can induce the apoptosis of endometrial stem cells.
