大肠湿热型溃疡性结肠炎患者Toll-like2/4受体mRNA表达及溃结灵的干预作用

摘    要:目的探讨溃结灵颗粒配合柳氮磺胺吡啶(SASP)对大肠湿热型溃疡性结肠炎(UC)的作用机制。方法活动期UC(大肠湿热证)患者随机分为治疗组(溃结灵+SASP)与对照组(SASP),以RT-PCR方法检测治疗后肠黏膜组织内的TLR2/4mRNA表达的变化。结果病变组的TLR2/4 mRNA表达较正常对照组明显增强。治疗组治疗后较治疗前TLR2/4 mRNA表达明显减少;对照组治疗后较治疗前TLR2 mRNA表达有所减少,但无显著性差异,TLR4 mRNA表达明显减少。治疗组与对照组TLR2 mRNA表达治疗后差值比较有显著差异。治疗组与对照组的TLR2/4 mRNA表达治疗后差值比较亦有显著差异。结论溃结灵配合SASP治疗很可能是通过抑止TLR4、TLR2 mRNA表达,减弱了CD14和TLR2的协同作用,抑制TLR2依赖的脂蛋白信号转导,阻断TLR4蛋白,进而磷酸化激活κB的功能减弱,使损伤的内皮细胞得以修复,而达到治疗UC的作用。

Changes of Toll-like Receptor 2/4 Gene Expression and Effects of Kuijieling Granules in the Patient of Ulcerative Colitis of Damp Heat in the Large Intestine Stagnation Type

Abstract:Objective: To study mechanism of Kuijieling Granules(KG) combined with SASP in treating ulcerative colitis(UC) . Methods: The patients of active UC (damp - heat syndrome) were randomly divided into 2 groups: KG + SASP group and SASP group. The TLR2 - 4mRNA expression in all cases were detected by RT - PCR. Results: There were obvious inereasement of the expression of TLR2- 4 mRNA in the pathological changes group. The TLR2 - 4mRNA expression were decreased by KG + SASP treatment . Conclusion: KG combined with SASP has definite effect on UC. it probably controls the gene expression of TLR2 - 4, singal conduction of BLP depend on by TLR2 and makes endothelial cells restored.
