小干扰RNA的设计及原理

摘    要:RNA干扰技术在人类基因功能研究、药物靶点鉴定等方面备受青睐,并且具有潜在的临床应用价值。但只有设计合理的小干扰RNA (siRNA)才能高效、特异地抑制靶基因表达。本文总结了siRNA序列的一般特点以及设计siRNA的方法和相应原理,并介绍了如何利用实验手段,验证RNA干扰的有效性及特异性。从而促进RNA干扰技术的广泛开展。

Abstract:RNA interference technique has been revolutionizing functional genomics, drug target identification and validation. And they are potentially valuable in clinical practice. Only well-designed siRNA can specifically and efficiently inhibit the expression of the target genes. This article reviews the features of siRNA sequences, principles and mechanisms of siRNA design, and the experimental strategies, testing of the efficiency and specificity of RNA interference, to provide information for the wider application of RNA interference technique.
