VEGFR-3抗体对人喉鳞癌裸鼠移植瘤模型的影响

摘    要:目的 研究血管内皮生长因子受体3(vascular endothelial growth factor receptor3,VEGFR-3)抗体对人喉鳞癌裸鼠移植瘤模型生长的抑制作用。方法 利用人喉鳞癌细胞株Hep-2细胞建立裸鼠喉癌模型。将24只裸鼠模型随机分成2组:治疗组和空白对照组,每组12只,分别腹腔内注射VEGFR-3抗体和生理盐水,观察肿瘤体积及重量变化,进行微淋巴管计数,绘制生长曲线并计算抑瘤率。结果VEGFR-3抗体组移植瘤体积和重量小于生理盐水组,有统计学意义(t体积=56.90,P〈0.05;t重量=11.64,P〈0.05)。EGFR-3抗体抑瘤率为48.21%。但组间微淋巴管计数无差异(t=1.48,P〉0.05)。结论 VEGFR-3抗体可以抑制裸鼠人喉鳞癌细胞移植瘤的生长,其作用机制可能与VEGFC和VEGFR-3的抑制有关。

Abstract:Objective To investigate the inhibitory effect of vascular endothelial growth factor receptor 3 (VEGFR-3) antibody on the transplantation model of human laryngeal squamous carcinoma in nude mice. Methods Human laryngeal carcinoma cells from Hep-2 cell strain were transplanted subcutaneously to BALB/c-nu/nu nude mice to establish the tumor model. Twenty-four nude model mice were randomized into two groups: treated group and control group, treated with VEGFR-3 antibody and isotonic Na chloride by intraperitoneal injection, respectively. The tumor volume, tumor weight and tumor inhibition rate were detected. Results Tumor volume and weight were significantly less in treated group than in control group (tvolume=56.90,P<0.05; tweight=11.64, P<0.05). The tumor inhibition rate in treated group was 48.21%. There was no significant difference in microlymphatic counts between two groups(t=1.48, P>0.05). Conclusion VEGFR-3 antibody could inhibit the growth of transplantation tumor of human laryngeal carcinoma in nude mice, and its mechanism may be associated with inhibitation of VEGFC and VEGFR-3.
