5,4’-二-正辛烷氧基-7-二氟亚甲基异黄酮体外抗人卵巢癌作用研究

摘    要:目的:研究5,4’-二-正辛烷氧基-7-二氟亚甲基异黄酮(DOdFMG)体外抗人卵巢癌作用。方法:体外培养CoC1细胞,MTT比色法测定DOdFMG对CoC1细胞增殖的影响;台盼蓝染色细胞计数法评价DodFMG对CoC1细胞的生长抑制作用;AO/EB染色法荧光显微镜观察DOdFMG诱导CoC1凋亡细胞的形态学改变;Western blotting法分析DOdFMG对CoC1细胞NF-κB蛋白表达和活性的影响。结果:MTT比色法结果显示,DOdFMG有效抑制CoC1细胞增殖活性,呈剂量依赖性;其IC50值为11.9μM。DOdFMG显著抑制CoC1细胞的生长,DOdFMG(10.0μM)使CoC1细胞群体倍增时间从37.8 h(溶媒对照组)延长到50.6 h;AO/EB染色荧光显微镜观察DOdFMG处理后,部分CoC1细胞呈现典型凋亡细胞形态特征;Western blotting分析结果表明:DOdFMG抑制NF-κB/p65表达,呈浓度和时间依赖性。结论:DOdFMG具有抑制体外培养人卵巢癌CoC1细胞增殖、生长和诱导细胞凋亡作用;其作用与抑制NF-κB/p65表达有关。

Abstract:Objective To investigate the antitumor effect 5,4'-Din-octoxyl-7-gem Difluoromethylene-genistein(DOdFMG) on human ovarian cancer cell in vitro.Methods CoC1cells was cultured in vitro.MTT assay was used to determine the effect of DOdFMG on the proliferation of CoC1 cells.The trypan blue exclusion method was used to investigate the growth inhibitory effect of DOdFMG in CoC1 cells.AO/EB fluorescence staining was used to observe the morphologic changes of apoptosis induced by DOdFMG in CoC1 cells.Western blot was used to analyze expression of NF-κB/p65 protein in CoC1 cells.Results DOdFMG significantly suppressed proliferation of CoC1 cells in a dose-dependant manner,and its IC50 was 11.9 μM.DOdFMG at 10.0μM could inhibit the growth of CoC1 cells and prolong CoC1 cell multiple time from 37.8h to 50.6h.Typical morphologic changes of apoptosis could be observed after treatment with DOdFMG by fluorescence microscope using AO/EB fluorescence staining.Western blotting indicated that expression of NF-κB/p65 protein decreased in a dose-and time-dependent manner.Conclusion DOdFMG could inhibit proliferation and growth and induce apotosis of human ovarian cancer CoC1 cells,and its mechanism might be associated with the inhibition of expression of NF-κB/p65 protein.
