D-半乳糖致衰老小鼠脾脏T细胞亚群的改变及其意义

摘    要:目的:探讨D-半乳糖所致亚急性衰老小鼠脾脏T细胞亚群的改变及其意义。方法:运用100 g/L D-半乳糖溶液建立小鼠亚急性衰老模型,并对此衰老模型进行生物学鉴定。ELISA检测模型小鼠血清中IFN-γ和IL-4的含量;流式细胞术检测模型小鼠脾脏中初始、活化及调节性T细胞相关分子的改变。结果:模型组小鼠血清中IFN-γ和IL-4含量分别为(82.93±2.74)pg/mL和(125.41±3.16)pg/mL,与对照组(125.41±3.16)pg/mL和(8.28±2.47)pg/mL]相比含量降低(P<0.01)。脾脏中初始T细胞相关分子CD45RA表达下降模型组:(4.46±1.21)%,对照组:(7.38±1.03)%,P<0.01];活化T细胞相关分子CD25表达下降模型组:(6.74±0.81)%,对照组:(5.14±1.25)%,P<0.05];Foxp3在CD4+CD25+T细胞中表达上升模型组:(18.32±1.44)%,对照组:(10.14±1.52)%,P<0.05]。结论:机体衰老时脾脏中初始和活化T细胞减少,CD4+CD25+Foxp3+调节性T细胞(Tr)增多,T细胞活化及免疫调节能力下降。

Abstract:AIM: To explore the changes and significance of spleen T cell subsets in ageing mice induced by D-galactose.METHODS: Ageing mice model was successfully established by 100 g/L D-galactose.The content of IFN-γ and IL-4 in serum was measured by ELISA.T cell subsets were detected by Immunofluorescence technique and flow cytometry.RESULTS: The content of IFN-γ and IL-4 in the serum was decreased of model group(P<0.01).The naive T cell related molecule,CD45RA was decreased(P<0.05).T cell activation-related molecule,CD25 was decreased(P<0.05).The Foxp3 in CD4+CD25+ T cell was increased(P<0.01).CONCLUSION: In spleen of the ageing mice,the percentage of naive and active T cell are decreased,but The percentage of CD4+ Tr subset is increased.
