卵巢恶性肿瘤p53基因5—7外显子突变及蛋白阳性的生物学意义

摘    要:目的比较研究卵巢恶性肿瘤p53基因第5,6,7外显子突变与p53基因蛋白的过度表达和临床生物学行为的相关性.方法对50例卵巢恶性肿瘤及10例良性肿瘤石蜡切片进行p53蛋白免疫组化检测,其中23例卵巢恶性肿瘤的新鲜标本进行了p53基因第5,6,7外显子PCRSSCP检测.结果50例卵巢恶性肿瘤中12例p53蛋白阳性表达24%(12/50).上皮癌29%(9/28)生殖细胞肿瘤10%(1/10)间质细胞瘤25%(2/8),转移瘤25%(1/4)在23例标本PCRSSCP检测中突变率为35%(8/23)第5,6,7外显子突变率分别为9%(2/23).9%(2/23).17%(4/23).两者均阳性者为2例,即PCR-SSCP检测和蛋白表达均阳性第6,7外显子各1例.结论①卵巢恶性肿瘤存在p53基因突变蛋白的过度表达,但突变与蛋白阳性表达并非完全一致;②p53基因突变及其蛋白过度表达与卵巢恶性肿瘤组织类型和临床分期无明显相关件.

Abstract:AIM: To study the telatlonship between the muta-tions of exons 5-7 in p53 gene and protein overexpression and clinical biological behavior of malignant ovarian tumors METHJODS:Immunohistochemical staining method was used to examine the Parafin embeuueu sections of 50 cases of malignant ovarian and tumors and 10 cases of benign tumors.Analysis of e-cons 5--7 of the p53 gene was performed in 23 o-varian malignance. RESULTS: Positive expression rate of p53for ovarian maligllancc was 24 % (12/50), epithelial cell type29%(8/28 ), genetic cell type 10% (1/10 ), interstitial celltype 25% (2/8 ), and metastatic tumors 25% (1/4 ), Amongthe 23 ovarian malignance subjected to PCR--SSCP test, therewere 35% (8/23), 9% (2/23) in exon 5, 9% (2/23) in exon6; 17% (4/23) in cxon 7. Two were posillve for both PCR-SSCP and immunostaining test. One was in exon 6, anotherwas in exon 7. CONCLUSION: Mutations of the p53 tu-mor suppressor gene have overexpression of p53 protein in o-varian malignance. but they do not coincide; Mutation ofthe p53 gene and overexprcssion of P53 protein have no obvi-ous reiations with pathological types and clinical stages.
