小剂量顺铂联合5-氟尿嘧啶对胃癌细胞增殖抑制及其机制的研究

摘    要:目的:探讨小剂量顺铂(DDP)、5-氟尿嘧啶(5-FU)在体外联合用药对SGC-7901胃癌细胞及胃癌耐药细胞株SGC-7901/5-FU细胞增殖及其相关基因表达的影响。方法:采用MTT法观察在小剂量DDP、5-Fu联合用药前后SGC-7901与SGC-7901/5-FU存活率的差异,同时应用免疫组化方法检测SGC-7901与SGC-7901/5-FU中p53、Fas/FasL基因的表达情况。结果:DDP与5-FU均有抑制肿瘤细胞增殖作用,且有浓度相关性;尤以DDP联合5-FU后对细胞存活率的影响明显高于两种药物的单独使用(P〈0.01),小剂量DDP联合应用5-FU时效果最明显,但对SGC-7901/5-FU细胞增殖抑制作用明显小于SHC-7901;小剂量联合DDP+5-FU,p53、FasL基因表达的阳性率显著高于单纯的5-FU组或DDP组(P〈0.01),而Fas基因表达则相反。结论:小剂量DDP联合5-FU对胃癌细胞具有明显抑制作用,对耐药株也有-定抑制作用,但效果弱于非耐药株。p53、Fas/FasL基因对胃癌细胞的增殖活性有重要作用,在小剂量DDP联合5-FU比各单药应用组基因的改变更明显(P〈0.01)。

Abstract:Objective: To evaluate the effect of 5 - FU combined with low - dose cisplatin on the expression of pro- liferation gene of SGC - 7901 gastric cancer cell and gastric cancerous resistance cell stain SGC - 7901/5 - FU. Methods:Inhibition effect of 5 -FU and low- dose cispiatin of different concentration to SGC -7901 cells and gas- tric cancerous resistance cell stain SGC -7901/5 -FU were examined by using MTY. The expression of p53, Fas/Fas L gene on the SGC -7901 gastric cell lines and SGC -7901/5 -FU was detected by immunocytology. Results: MTY assay results showed that 5 - FU and DDP can obvious affect carcinoma cells proliferation and concentration revel- ance. DDP combined with 5 - FU on the cell survival rate was significantly higher than drug used alone (P 〈0.01 ), the effect was most obvious when combined with low - dose DDP. Immunocytochemistry results showed that the posi- tive rate of p53, Fas L gene expression in 5 - FU + DDP group was significantly higher than 5 - FU group and DDP group( P 〈 0. O1 ) ,but Fas gene expression was just opposite. Conclusion:5 - FU combined with low - dose cisplatin can inhibit and kill the gastric cancer cell ,which also has a certain inhibitory effect on drug - resistant strains. The expression of p53, Fas/Fas L gene, are very important to the proliferation of gastric cancer cell, and obviously changed by 5 - FU combined with low - dose cispiatin.
