摘    要:目的研究Survivin特异性siRNA对人结肠癌细胞株HCT116增殖和对化疗药物奥沙利铂敏感性的影响。方法体外将Survivin特异性siRNA表达载体pgsiRNA-Survivin转染HCT116细胞,通过Western blot和Real time-PCR方法检测细胞Survivin的表达,采用流式细胞术分析细胞周期分布,MTT法检测干扰Survivin后HCT116细胞对奥沙利铂的敏感性。结果 pgsiRNA-Survivin转染HCT116细胞48 h后,细胞的Survivin基因表达明显降低,导致了HCT116细胞G2/M期阻滞;pgsiRNA-Survivin转染组细胞对奥沙利铂的敏感性显著性增强。结论 Survivin特异性siRNA能显著沉默HCT116细胞Survivin基因,抑制细胞增殖,并增强HCT116细胞对奥沙利铂的敏感性。
