摘    要:目的:探讨羟基磷灰石(Hydroxyapatite,HA)压片材料对小鼠MC3T3成骨细胞凋亡的影响,提供一种方便可行的组压片材料的生物相容性检测方法。方法:小鼠MC3T3成骨细胞株接种在HA压片材料上,应用光学显微镜和PI-Hoechst33342双染色荧光显微镜观察细胞,采用RT-PCR方法分析HA对Id2(Inhibitor of differentiation 2,分化抑制因子2)和OPN(osteopontin,骨桥蛋白)等基因表达的影响。结果:HA压片材料组MC3T3细胞凋亡率(78.7%)显著高于正常对照组(24.6%)(t’=28.1,P=0.000)。HA压片材料双荧光染色可以直接检测种子细胞的凋亡。结论:HA压片可促进小鼠MC3T3成骨细胞凋亡。

Abstract:Objective: To investigate the effect of hydroxyapatite(HA) material on apoptosis of mouse MC3T3 cells, and find a convenient and feasible method for the detection of biocompatibility about HA material. Methods: Mouse MC3T3 cells were cultured on the HA disks, dyed with PI-Hoechst33342 and observed under light microscope and fluorescence microscope. Expression of DNA of Id2 (Inhibitor of differentiation 2) and OPN (osteopontin) was tested through RT-PCR. Results: The rate (78.7%) of apoptosis of mouse MC3T3 cells cultured on HA disks was significantly higher than the rate (24.6%) of the control groups. (t'=28.1, P=0.000). The results implied that double fluorescent dyeing can detect apoptosis of seed cells directly. Conclusions: HA material can promote apoptosis of MC3T3 cells.
