加味丹参饮及其拆方对BMSCs移植IRI鼠心肌ang-1表达的影响

摘    要:目的:观察加味丹参饮及其拆方干预骨髓干细胞(bone mesenchymal stem cells,LBMSCs)移植后心肌缺血再灌注损伤(IRI)模型大鼠,比较其对梗死区及梗死边缘心肌血管生素-1(angiogenin-1,ang-1)蛋白表达的影响。方法:30只幼鼠用以提取BMSCs,采用全骨髓贴壁法培养、传代、扩增,观察细胞形态,传至P3时,用以移植。300只大鼠随机分为假手术组、模型组、加味丹参饮组、丹参饮组、加味组。对除假手术组外的其余组大鼠均进行心肌IRI造模,再分别在大鼠心肌梗死区边缘移植BMSCs细胞培养液。术后14d取心肌进行HE染色;术后3、7、14d取心肌组织用免疫组化法检测ang—1蛋白的表达。结果:免疫组化结果显示.与假手术组比较,手术组ang—1的阳性表达均升高(P〈0.05);与模型组比较,用药组ang—1阳性表达均升高,其中以加味丹参饮组效果最佳(P〈0.05);丹参饮组与加味组比较,差异无统计学意义(P〉0.05)。结论:加味丹参饮及其拆方对IRI大鼠心肌具有增加ang-1蛋白表达、改善梗死区心肌组织病理学形态的作用,且加味丹参饮的作用明显优于拆方。提示加味丹参饮及其拆方对缺血心肌的保护作用可能与增加ang-1的阳性表达有关,且加味丹参饮中药物的配伍可使其功用发生协同促进作用。

Effects of Jiawei Danshen decoction and its components on myocardial angiogenin - 1 expression in rats with ischemia - reperfusion injury after transplantation of bone marrow mesenchymal stem cells

Abstract:Objective :To observe the effects of Jiawei Danshen decoction and its components on the protein expression of angiogenin - 1 ( ang - 1 ) in infarct zone and infarct border zone among rats with myocardial ischemia - reperfusion injury (IRI) after transplantation of bone marrow mesenchymal stem cells (BMSCs). Methods : Thirty young rats were selected for isolation of BMSCs. Whole bone marrow adherent culture, subculture, and multiplication, as well as observation of cell morphology, were performed. The cells of the third generation were harvested for transplantation. A total of 300 rats were randomly divided into sham -operation group, model group, Jiawei Danshen decoction group, Danshen decoction group, and Jiawei group. All rats except those in sham - operation group were treated to establish a myocardial IRI model. Culture solution of BMSCs was transplanted into the infarct border zone of myocardium. At 14 d after operation,the myocardium was harvested for HE staining. At 3,7, and 14 d after operation,myocardial tissues were harvested,and the protein expression of ang - 1 was measured by immunohistoehemistry. Resuhs : Compared with the sham - operation group, other groups had sig- nificantly increased ang - 1 expression (P 〈 0. 05 ). Compared with the model group,the Jiawei Danshen decoction group, Danshen decoction group,and Jiawei group had significantly increased ang - 1 expression,and the Jiawei Danshen decoction group had the best outcome ( P 〈 0. 05 ) ; there was no significant difference between the Danshen decoction group and Jiawei group ( P 〉 0. 05 ). Conclusion : Jiawei Danshen decoction and its components can increase the protein expression of ang - 1 and reduce pathological changes in the infarct myocardium among rats with IRI, and Jiawei Danshen decoction has better efficacy than its components. It suggests that the protective effect of Jiawei Danshen decoction and its components may be related to increasing ang - 1 expression, and the components of Jiawei Danshen decoction show a synergistic effect.
