摘    要:目的:观察大豆异黄酮(soybean isoflavones)干预MMTV-erbB-2转基因小鼠,对乳腺癌组织中细胞外信号调节激酶1/2(extracellular signal regulated kinase 1/2,ERK1/2)和c-Fos蛋白表达的影响及意义.方法:选择鼠龄3周的健康MMTV-erbB-2转基因雌性小鼠100只,随机分为4 组(早期干预组、晚期干预组、全程干预组和对照组),观察各组小鼠乳腺癌的发病率和潜伏期,采用免疫组织化学法检测各组小鼠乳腺癌组织及正常乳腺组织中ERK1、ERK2和c-Fos蛋白的表达并进行统计学分析.结果:早期干预组、晚期干预组、全程干预组和对照组雌性MMTV-erbB-2转基因小鼠乳腺癌的发病率分别为48%、28%、20%和92%;与对照组比较,早期干预组、晚期干预组和全程干预组发病率均降低(P<0.01);全程干预组发病率低于早期干预组(P<0.05),晚期干预组与早期干预组和全程干预组比较,发病率差异均无统计学意义;各组小鼠乳腺癌潜伏期分别为(47.67±9.25)、(47.29±14.13)、(51.8±6.69)和(49.91±8.07)周,各组间差异均无统计学意义.各组小鼠的肿瘤直径比较差异无统计学意义(P>0.05);ERK1、ERK2和c-Fos在乳腺癌组织中的阳性表达率分别为71.6%、68.7%和61.2%,在小鼠正常乳腺组织中阳性表达率分别为27.7%、19.1%和12.8%,差异有统计学意义(P<0.01);与对照组相比,早期干预组、晚期干预组和全程干预组乳腺癌组织ERK1、ERK2和c-Fos阳性表达均有所降低,但差异无统计学意义(P>0.05).结论:大豆异黄酮对MMTV-erbB-2转基因雌鼠乳腺癌的发生具有一定的预防作用,丝裂原活化蛋白激酶(mitogen activated protein kinase,MAPK)信号转导通路在乳腺癌的发生、发展过程中可能起着重要作用.
