糙叶败酱总木脂素对K562细胞体外生长的影响

摘    要:目的:探讨糙叶败酱提取物总木脂素对K562细胞体外生长的影响及作用机制。方法:采用四甲基偶氮唑盐比色法(monotetrazolium test,MTT)测定糙叶败酱总木脂素对K562细胞生长的影响;采用电镜技术对其进行形态学观察;采用流式细胞仪对其进行DNA含量分析,检测经药物作用后的细胞凋亡率。结果:糙叶败酱总木脂素对K562细胞生长有明显的抑制作用,抑制作用随药物浓度增加及作用时间延长而有加强趋势,细胞经糙叶败酱总木脂素作用96h,IC5021.16μg/mL。电镜下观察到经糙叶败酱总木脂素作用48h后,一部分K562细胞体积变小,胞浆浓缩,胞核染色质聚集或边集,核碎裂,凋亡小体形成,呈典型的凋亡形态特征。流式细胞仪可检测到经糙叶败酱总木脂素诱导K562细胞48h后产生明显的亚二倍体峰,细胞凋亡率为10.1%。结论:糙叶败酱总木脂素可抑制K562细胞的生长,作用的机制与其诱导K562细胞凋亡有关。

Abstract:Objective:To discuss the effects of Patrinia scabra Bunge lignanoid on K562 cells growth in vitro and the relative mechaniam.Methods:The effects of Patrinia scabra Bunge lignanoid on K562 cells growth in vitro were assayed by monotetrazolium test(MTT).In this research,transmission electron microscope(TEM) was employed to observe the apoptotic rates of K562 cells treated with Patrinia scabra Bunge lignanoid,and flow cytometry(FCM) was used to analyze the DNA content and to detect the apoptotic peak(AP).Results:Patrinia scabra Bunge lignanoid had an effect on K562 cells in a concentration-and-time-dependent manner.TEM test showed that 48 hours after treated with Patrinia scabra Bunge lignanoid,some K562 cells became smaller in size,and had the following typical features of cell apoptosis such as condensed cell nucleus,assembled chromatin,and disintegrated nucleus body.The apoptosis rate was 10.1%.Conclusion:Patrinia scabra Bunge lignanoid can inhibit the growth in vitro of K562 cells and the mechanism is that it can induce the apoptosis of K562 cells.
