摘    要:具有抗8-AG和HGPRT~-遗传标记的HL-60-AR细胞与维生素A酸或二甲基亚砜一起保温后,细胞对NBT还原反应能力增强;细胞膜表面出现C_2补体受体;细胞形态按粒系白细胞途径向较成熟的方向发育。同时,细胞并合~3H-TdR的速率降低;细胞几乎全部丧失在软琼脂中形成集落的能力。HL-60-AR细胞对次黄嘌呤的诱导分化反应不敏感。

Abstract:A human promyelocytic leukemia cell mutant (HL-60-AR) with characteristics of resistence to 8-AG and deficiency in HGPRT has been established from HL-60 cell line in our institute. In this paper, potency of induced differentiation of this mutant cell line was studied with treatment in Vitro of retinoic acid, dimethyl salfoxide and hypoxanthine.Results indicate that when HL-60-AR cells are incubated with retinoic acid (10-6mol) or dimethyl sulfoxide (1.4%) for 6 days, the ability of the cells to reduce NBT dye is obviously increased with while C3 complement receptors demonstrable in cell membranes.It was found that the treated cells assumed phenotypes of di-ffereatiated myeloid cells, showing both decrease of nucleus-cytolasmic ratio and 3H-TdR incorporation rate. Almost all of the treated cells lose the capacity of forr. ng colony in soft agar medium. However, probably due to deficiency in HGPRT.no inductive effect can be observed in HL-60-AR mutant cells following hypoxanthine treatment.
