Rb基因和PCNA在前列腺癌中表达的意义及相关性分析

摘    要:目的 探讨视网膜母细胞瘤易感基因 (Rb)和增殖细胞核抗原 (PCNA)在前列腺癌中表达的意义及其相互关系 .方法 采用免疫组织化学 SABC法对 36例前列腺癌标本 Rb基因和 PCNA进行检测 ,并应用图像分析仪及统计学方法对两者在不同病理分级及临床分期中的表达进行对比及相关分析 .结果  36例前列腺癌标本中 Rb表达阳性 17例 ,阳性率47.2 % ,其表达在前列腺癌的不同病理分级及临床分期之间有显著差异 (P <0 .0 5 ) ;PCNA在标本中的表达强度在不同病理分级及临床分期之间有显著差异 (P<0 .0 5 ) ;同时还发现 Rb表达与 PCNA表达存在负相关 ,即 Rb阳性标本中 PC-NA表达弱而 Rb阴性标本中 PCNA表达强 ,两者比较有显著性差异 (P<0 .0 5 ) .结论  Rb基因参与了前列腺癌的发生、发展过程 ,在发病机制中可能涉及到 Rb和 PCNA调节通路的异常 ,检测 Rb基因和 PCNA有助于前列腺癌恶性程度的判定及预后 .

Abstract:AIM To discuss the expression and correlation of retinoblastoma susceptibility gene and proliferating cell nu clear antigen in the prostatic cancer. METHODS SABC immunohistochemistry was used to determinate Rb gene and PCNA in 36 samples of the prostatic cancer. The image pattern analyzer and the statistics methods were used to do the comparative analysis of Rb gene and PCNA in the different pathological grading and clinical phases of the prostatic cancer. RESULTS immunohistochemistry staining was Rb positive in 17 of 36 samples,rated 47.2%. The expression of Rb had significant differences in different pathological grading and clinical phases ( P <0.05). The expression of PCNA also had significant differences in different pathological grading and clinical phases ( P <0.05). The expression of PCNA was weak in the Rb positive expression samples and was strong in the Rb negative expression samples. There was a significant difference between Rb and PCNA expression ( P <0.05). CONCLUSION Rb is involved in the occurrence and development of the prostatic cancer. As for pathological mechanism, it is related to the abnormality of Rb and PCNA regulating path. The determination of Rb and PCNA can be an index for estimating the level of malignant and progression of the prostatic cancer.
