两种rHuIL-6/GM-CSF融合蛋白对化疗药物所致小鼠白细胞减少的恢复作用

以BALB/c系小鼠作为实验动物 ,探讨两种rHuIL 6 /GM CSF融合蛋白 包括IL 6 (1 184 )GM CSF (9 12 7) (简称IG1) ,IL 6 (2 4 184 ) GM CSF (9 12 7) (简称IG2 ) ]在小鼠体内的生物学活性 ,观察它们对环磷酰胺所致BALB/c系小鼠白细胞减少及造血功能的影响。注射环磷酰胺建立化疗模型 ,分别同时注射融合蛋白 7d ,于 13d摘除眼球采血 ,并进行外周血白细胞记数 ,骨髓有核细胞 ,单个核细胞记数及CFU GM的测定。实验结果表明两种rHuIL 6 /GM CSF融合蛋白对环磷酰胺所致小鼠白细胞减少及造血功能的损伤具有不同程度的修复作用。

Effects of Two rHuIL-6/GM-CSF Fusion Protein on the Recovery of the Chemotherapy Drug Induced Leukocytopenia in Mice

Using BALB/c mice as the experimental model to evaluate the in vivo biological activities of two fusion proteins including.IL-6 (1-184)-GM-CSF (9-127) (IC1) and IL-6 (24-184)-GM-CSF (9-127) (IG2),and to observe their influences on the recovery of the cyclophosphamide-induced leukocytopenia in mice, the chemotherapy model of mice was established by injections of cyclophosphamide, and was daily injected subcutaneously with the fusion proteins for 7 days. Blood was taken from orbital bulb for the WBC count of the peripheral blood, and bone marrow was taken to determine the counts of bone marrow nucleated cells and monocytes as well as for the determination of the CFU-GM. The experimental results showed that these two fusion proteins had different degree of modulating effects on the recovery of the cyclophosphamide-induced leukocytopenia and the damages of the hematopoietic cells in BALB/c mice.
