中国汉族、蒙古族和泰国人群中D8S1132基因座的遗传多态性研究

摘    要:目的 获得D8S1132基因座的群体遗传学数据并分析其基因频率在不同群体间是否有差异。方法 采用PCR、聚丙烯酰胺凝胶电泳及银染技术分析中国成都地区汉族、海拉尔蒙古族和泰国曼谷地区泰国人群三个群体中D8S1132基因座的遗传多态性,获得三个群体D8S1132基因座的群体遗传学数据。结果 从380份分别采自成都地区汉族、海拉尔蒙古族和曼谷泰国人三个群体的无血缘关系个体的静脉血,共发现11个等位基因,观测到42种基因型.观测杂和度为85.0%~89.5%,个人识别机率为94.1%~95.5%,观测到的基因型频率分布符合Hardy-Weinberg平衡定律。等位基因频率的分布在三个群体间有显著差异。结论 D8S1132基因座个人识别能力高,方法简便、灵敏,重复性好,在法医学个人识别和亲子鉴定应用中有较高价值。

Abstract:Objective The aim of the present study was to obtain the population genetic data of short tandem repeat(STR) locus D8S1132 and study the difference of allelic frequency distributions among different populations. Methods A sample of 380 unrelated individuals from 3 populations (Han, Mongolian, Thai) were studied using single PCR followed by polyacrylamide gel electrophoresis and silver staining. Results Eleven alleles and 42 genotypes were observed. The observed heterozygosity and the discrimination power were 85.0%-89.5% and 94.1%-95.5%, respectively. The genotype distributions of D8S1132 locus in 3 populations were in accordance with Hardy-Weinberg equilibrium and the difference of allelic frequency distributions among 3 populations was obvious. Conclusion The methods established are easy-to-perform and sensitive. The discrimination power and exclusion chances of this locus are desirable for forensic analysis.
