积雪草苷对小鼠脂多糖致痛觉增敏及对MPO、NO和TNF-α影响研究

摘    要: 目的观察积雪草苷(aisaticoside,AS)对小鼠腹腔注射脂多糖(lipopolysaccharide,LPS)诱导痛觉增敏型疼痛的影响,并探讨AS对髓过氧化物酶(myeloperoxidase,MPO)、一氧化氮(nitrogen monoxidum,NO)和α-肿瘤坏死因子(α-tumour necrosis factor,TNF-α)的影响。方法随机将小鼠分为空白组、模型组、盐酸哌替啶组、AS5,15,45mg·kg-1 6组,通过腹腔注射LPS100μg·kg-1建立痛阈增敏型疼痛模型,采用醋酸扭体法、热板法观察AS的镇痛效应;通过相关试剂盒检测小鼠脑组织MPO和NO,ELISA法检测血浆TNF-α含量的变化。结果AS 5,15,45mg·kg-1各组明显减少醋酸扭体次数,提高小鼠痛阈。降低脑组织MPO活性、NO含量和血浆TNF-α含量。结论AS具有明显镇痛效应,其镇痛效应可能与降低MPO、NO和TNF-α有关。

Abstract:OBJECTIVE To observe the analgesia effect of aisaticoside on hyperalgesia induced by intraperitoneal LPS in mice,and investigate the change of MPO,NO and TNF-α.METHODS Mice were divided randomly into normal group,model group,spasmedal group and AS 5,15,45 mg·kg-1 group,hyperalgesia was induced by intraperitoneal administration of LPS 100 μg·kg-1 in mice.The analgesia effect of AS was assessed by the hot-plate stimulation and the writhing test induced by acetic acid,the activity of MPO,NO in brain tissue was tested by kits;the content of TNF-α in plasma was tested by ELISA.RESULTS AS doses of 5,15,45 mg·kg-1 significantly reduced the number of writhing,increased pain threshold,decreased the activity of MPO,NO and TNF-α.CONCLUSION AS have the effect of analgesia and its effect maybe related with inhibiting the MPO,NO and TNF-α.
