摘    要:目的筛选食管鳞状细胞癌(esophageal squamous cell carcinoma,ESCC)的免疫和预后相关特征基因。方法基于单样本基因集富集分析(ssGSEA),将TCGA数据库中的ESCC序列和临床资料分为3个免疫浸润组。应用表达数据(ESTIMATE)算法对ESCC组织中的基质细胞和免疫细胞进行分组效应评估,并应用加权基因共表达网络分析(WGCNA)筛选与免疫表型相关的共表达基因模块,挑选最相关模块中的节点基因,分析其与患者预后的相关性。结果利用ssGSEA算法,81例肿瘤样本分为高免疫组(14例)、中等免疫组(46例)和低免疫组(21例),并用HLA-I和PD-L1基因验证了分组效果。利用WGCNA获得了与免疫相关的模块,在此模块中找到6个免疫相关hub基因,其中HAVCR2 mRNA与ESCC患者生存相关。结论HAVCR2作为免疫相关基因,可能参与了ESCC的发生、发展,是ESCC潜在预后标志物。

Abstract:Objective To screen the immune-and prognostic-related signature of esophageal squamous cell carcinoma(ESCC).Methods ESCC sequencing and clinical data from TCGA database were clustered into three immune infiltration groups based on single sample gene set enrichment analysis(ssGSEA).Estimation of Stromal and Immune cells in Malignant Tumor tissues using Expression data(ESTIMATE)algorithm was performed to assess grouping effects,and weighted gene co-expression network analysis(WGCNA)was performed to screen the co-expressed gene modules related to the immune phenotype.Hub genes was selected from immune-related modules.Results Using ssGSEA algorithm,81 tumor ESCC samples were divided into high immune group(14 cases),medium immune group(46 cases)and low immune group(21 cases).Six hub genes were screened out from immune-related module acquired by WGCNA,in which HAVCR2 mRNA was related to survival of ESCC patients.Conclusion HAVCR2 might be involved in the carcinogenesis and be prognosis markers of ESCC.
