摘    要:目的 :本法建立了高效液相色谱法 (HPLC)同时测定抗癫痫药苯巴比妥 (PB)、苯妥英钠 (DPH)及卡马西平(CBZ)的血药浓度。方法 :将样本在PH7.6条件下用二氯甲烷提取后经纯净的空气在 5 0℃水浴中吹干 ,再用甲醇重溶后进行色谱分析。色谱条件 :ELITEC18柱 ,流动相 :甲醇 -水 (6 0∶4 0 ) ,流速 :1mL/min ,柱温 :室温 ,检测波长 :2 35nm。结果 :PB、DPH、CBZ的保留时间分别为 3.5 5min、5 .89min、7.14min ,最低检出浓度分别为 1.2 1ug/ml、0 .79ug/ml和 0 .4 4ug/ml。在三个不同浓度下 ,PB、DPH、CBZ的平均回收率各为 10 0 .18%、98.32 %和 10 0 .0 6 % ,日内和日间变异均低于 9.87% ,其他常用抗癫痫药物未见干扰 ,在分析范围内具有良好的线性关系。结论 :该方法操作简便、快捷、准确 ,可满足临床快速监测的需要

Abstract:Objective: To establish the method to determine some anti-epilepsy drugs PB,DPH and CBZ concentration in the blood by HPLC. Methods: Under the circumstance of PH 7.6, the samples were extracted by dichloromethane, and dried in the bath of 50℃ water by the pure air, then they were dissolved by methanol and analyzed by chromatography with the conditions: ELITEC C 18; mobile phase: methanol-water (60∶40); flowing rate: 1mL/min; column temperature: room; wave length:275nm. Results: The retaining time of PB,DPH and CBZ was 3.55min, 5.89min, 7.14min,the lowest concentration that can be detected was 1.21ug/ml, 0.79ug/ml and 0.44ug/ml. With the 3 different concentration, the average of the recovery rates were 00.18%,98.32% and 100.06% respectively. The difference between the day and night was averagely lower than 9.87%, the interference of other anti-epilepsy medications was unseen, the analyzed scope had the better linear relation. Conclusion: This method is quite simple, rapid and accurate, it is suitable to be used in determining the concentration of anti-epilepsy in human plasma.
