| 摘 要： | 建立一种体外筛选抗ＣＭＶ药物的ｉｓＥＬＩＳＡ法，方法 采用ＥＬＩＳＡ测定后培养细胞上的病毒抗原，并与空斑减少试验（ＰＲＡ）法进行比较。结果 ｉｓＥＬＩＳＡ法病毒接种量范围宽，重复性好，结果客观可靠，操作简便，保需５ｄ即可观察结果。结论：ＥＬＩＳＡ可代替ＰＲＡ法用于体外抗ＣＭＶ药物的筛选。

Application of in situ ELISA(isELISA) in anti-cytomegalovirus(CMV) compound screening |

| Abstract: | Purpose To establish an isELISA as an improved procedure for anti CMV susceptibility testing.Methods CMV antigens were detected on fixed virus infected cells and the method was evaluated by compared with plaque reduction assay(PRA),a standard method,on anti CMV compounds of ganciclovir and antisense oligodeoxynucleotide phosphorothioates(AS ODN).Results The ED 50 obtained by isELISA were similar with that by PRA.The isELISA is reproducible,relatively simple to perform,has a more objective interpretation,and can be completed in 5 days postinfection.Conclusions isELISA can be used to replace PRA for the evaluation of effect of anti CMV compounds. |
