南方红豆杉枝叶中黄酮组分的含量测定及抗氧化活性研究

摘    要:目的考察南方红豆杉枝叶乙醇提取物不同萃取组分中黄酮的含量及体外抗氧化作用。方法南方红豆杉枝叶乙醇提取物经过滤、浓缩、石油醚除杂后,分别用乙酸乙酯、正丁醇进行萃取,萃取后水混悬液部分用60%乙醇溶液定容,得到3种不同黄酮组分(TMA,TMB,TMC),采用分光光度法测定3种组分中黄酮的含量;通过1,1-二苯基-2-苦肼基(DPPH)清除率、还原力、总抗氧化活性3个体外抗氧化试验模型,研究并比较不同组分的抗氧化作用差异。结果南方红豆杉TMA,TMB,TMC中黄酮含量分别占总黄酮含量的50.17%,38.25%,11.58%。其清除DPPH自由基的能力、总抗氧化活性和还原力均随总黄酮质量浓度的增加而增强,3种组分抗氧化能力与所含黄酮的含量有一定的相关性。结论南方红豆杉3种黄酮组分均具有较强的抗氧化能力,对体外自由基有较好的清除作用。

Abstract:Objective To investigate the antioxidant activity and determination of different flavonoids components in branches and leaves of Taxus mairei(Lemee et Levl.) S.Y.Hu ex Liu in vitro.Methods The ethanol extracts in branches and leaves of Taxus mairei(Lemee et Levl.) S.Y.Hu ex Liu were handled through filter,concentration and petroleum ether impurity removal,and respectively using ethyl acetate,n-butanol for extraction.After extracting,used 60% ethanol solution to 100 mL capacity,and got three kinds of flavonoids components(TMA,TMB,TMC).Then the contents of total flavonoids of three different flavonoids components were determined by spectrophtometry.The antioxidant activity of different flavonoids components in vitro was evaluated by the methods of detecting DPPH clearance rate,reducing power and total antioxidant activity.Results The contents of total flavonoids in different extracts(TMA,TMB,TMC) of Taxus mairei(Lemee et Levl.) S.Y.Hu ex Liu were 50.17%,38.25%,11.58% respectively.The scavenging capacity of DPPH,reducing power and total antioxidant activity enhanced with increasing of mass concentration of total flavonoids.The antioxidant capacity of three extracts was associated with the content of total flavonoids.Conclusion The three flavonoids components from Taxus mairei(Lemee et Levl.) S.Y.Hu ex Liu have strong antioxidant capacity and better scavenging effect of free radical in vitro.
