| 摘 要： | 探讨蕲蛇酶溶栓作用机制。方法应用发色底物（Ｓ２３９０）显色法测定大鼠静脉注射蕲蛇酶２和４Ｕ／ｋｇ（３００和６００μｇ／ｋｇ）后５，１５，２５和４５ｍｉｎ血浆中组织型纤溶酶原激活剂（ｔ－ＰＡ）和纤溶酶原激活剂抑制物（ＰＡＩ－１）的活性。结果正常大鼠静脉注射蕲蛇酶后５ｍｉｎ血浆中ｔ－ＰＡ活性即开始升高，１５ｍｉｎ时达到高峰，低和高剂量组ｔ－ＰＡ活性（ＩＵ／ｍｌ）分别由给药前１６１．８１±２８．７８和１８９．１４±７１．３０升至３８９．５６±１４３．１０和４８２．８７±８５．６７，随后活性下降，至４５ｍｉｎ时基本恢复正常。与此同时，ＰＡＩ－１的活性变化不明显。结论蕲蛇酶溶栓作用与其促进ｔ－ＰＡ释放有关。

| Abstract: | Objective To study the mechanism of thrombolysis of thrombinlike enzymes from Agkistrodon acutus. Methods A chromogenic assay technique was used to identify the activity of tissue plasminogen activator(t PA) and tissue plasminogen activator inhibitor(PAI)in rat plasma. Results A high thrombin like enzymes(TLEs) increase of t PA activity was present after intravenous injection, but fell to a normal level in 45 min. The change of activity of PAI 1 was not significant. Conclusion The results indicated that t PA was released after intravenous injection of TLEs. It was relative to the thrombolytic action of TLEs. |
